circPTK2 siRNA is a small interfering RNA designed to specifically target and silence the expression of circPTK2, a circular RNA that is upregulated in bladder cancer. Mechanistically, circPTK2 acts as an oncogenic scaffold that binds to Poly(A) Binding Protein Cytoplasmic 1 (PABPC1), which in turn stabilizes the mRNA of SET Domain Bifurcated Histone Lysine Methyltransferase 1 (SETDB1). This stabilization leads to the overexpression of SETDB1, promoting epithelial-mesenchymal transition (EMT), enhancing tumor cell migration and invasion, and inducing resistance to gemcitabine. By utilizing the RNA interference (RNAi) pathway to degrade circPTK2 transcripts, this siRNA disrupts the circPTK2/PABPC1/SETDB1 signaling axis, thereby suppressing metastatic potential and restoring chemosensitivity in preclinical bladder cancer models.
