Overview

CPT (Circularly Permuted TRAIL) is a recombinant human TRAIL variant developed by Beijing Shadong Biotechnology. It is a modified version of the tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) where the N- and C-termini have been rearranged (circularly permuted) to enhance stability and pro-apoptotic activity. CPT functions as an agonist for death receptors DR4 (TRAIL-R1) and DR5 (TRAIL-R2), which are often overexpressed on the surface of cancer cells. Upon binding, CPT triggers the extrinsic apoptotic pathway, leading to the activation of caspases and subsequent cell death. It has been primarily evaluated in Phase 3 clinical trials for the treatment of relapsed or refractory multiple myeloma in combination with thalidomide and dexamethasone.
