Overview

Cirmtuzumab is a humanized monoclonal antibody that targets receptor tyrosine kinase-like orphan receptor 1, an oncoembryonic cell-surface receptor expressed on multiple hematologic malignancies and some solid tumors. It was developed by Oncternal Therapeutics after licensing from the University of California San Diego, and later sold with other ROR1 assets to Ho'ola Therapeutics in 2025. The antibody is designed to block Wnt5a-ROR1 signaling, thereby inhibiting downstream pathways linked to tumor cell proliferation, stemness, migration, and survival, including Rho GTPase, NF-kappaB, and STAT3 signaling. Clinical and investigator-sponsored development has included chronic lymphocytic leukemia, mantle cell lymphoma, relapsed or refractory chronic lymphocytic leukemia in combination with venetoclax, breast cancer in combination with paclitaxel, and combination studies with ibrutinib in chronic lymphocytic leukemia and mantle cell lymphoma.
