Overview

CIS-0602K is a mitochondrially directed insulin sensitizer being developed by Cirius Therapeutics for the treatment of type 2 diabetes. It is designed to replicate the insulin-sensitizing pharmacology of first-generation thiazolidinediones like pioglitazone while avoiding direct PPARγ-mediated side effects. The drug works by targeting the mitochondrial pyruvate carrier (MPC) to modulate metabolic pathways. Clinical studies have evaluated doses of 125 mg and 250 mg daily in patients with type 2 diabetes, demonstrating improvements in HbA1c and insulin-stimulated glucose uptake. It is also being explored in combination with incretin-based weight loss therapies like tirzepatide to manage weight regain and synergistically improve metabolic health.
