Overview

cis-cyclo(L-Phe-L-Pro) is a cyclic dipeptide (2,5-diketopiperazine) composed of L-phenylalanine and L-proline in the cis configuration. It is a secondary metabolite produced by various microorganisms including Lactobacillus plantarum and certain fungi. This compound exhibits broad-spectrum antibacterial and antifungal activities, with demonstrated efficacy against multidrug-resistant bacteria such as methicillin-resistant Staphylococcus aureus (MRSA), Enterococcus raffinosus, and also shows activity against influenza A (H3N2) virus[2][5]. Mechanistically, it interferes with bacterial quorum sensing systems—specifically N-acylhomoserine lactone-mediated signaling—and inhibits biofilm formation by affecting the expression of genes related to quorum sensing and polysaccharide intercellular adhesion[1][4]. Additionally, it displays free radical scavenging properties in vitro. Its precise molecular mechanism remains incompletely characterized but involves modulation of microbial communication pathways.
