CIS-DPI is a novel dry powder formulation of cisplatin designed for inhalation. Developed by InhaTarget Therapeutics, it enables local pulmonary delivery of the platinum-based chemotherapy agent cisplatin directly to lung tissue. This approach aims to maximize antitumor activity in the lungs while minimizing systemic toxicities commonly associated with intravenous administration. The mechanism of action is consistent with that of cisplatin—DNA cross-linking and inhibition of DNA synthesis—leading to apoptosis in rapidly dividing tumor cells. Preclinical studies have shown that CIS-DPI demonstrates dose-dependent antiproliferative effects in lung tumor models and can enhance immune responses when combined with immune checkpoint inhibitors such as anti-PD1 antibodies. Clinical development is focused on non-small cell lung cancer (NSCLC), particularly as an adjunct to standard-of-care immunotherapy or chemotherapy regimens[1][2][4][5][6].
