Overview

cis-Nerolidol is a naturally occurring sesquiterpene alcohol and a geometric isomer of nerolidol found in various essential oils, such as neroli and jasmine. It is currently under investigation for its potent antitumor properties, particularly in the context of bladder carcinoma. The compound exerts its effects by inducing DNA damage and endoplasmic reticulum (ER) stress. Mechanistically, it activates a signaling axis involving cyclic adenosine monophosphate (cAMP), calcium (Ca2+), and mitogen-activated protein kinase (MAPK) pathways. This activation leads to two distinct cell death pathways: an early caspase-dependent mechanism characterized by membrane blebbing and cell swelling, and a late caspase-independent mechanism involving cytoplasmic vacuolization. Beyond its oncology potential, cis-nerolidol exhibits antioxidant, antibacterial, and antiparasitic activities.
