Overview

CISH inactivated TIL refers to a cell therapy product consisting of autologous tumor-infiltrating lymphocytes (TILs) that have been genetically engineered ex vivo to inactivate the cytokine-inducible SH2-containing protein (CISH) gene using CRISPR/Cas9 technology. CISH is an intracellular immune checkpoint and negative regulator of T-cell receptor signaling. Inactivation of CISH enhances the anti-tumor activity and functional avidity of T cells by removing this inhibitory pathway, resulting in improved recognition and killing of cancer cells. This therapy is being developed primarily for solid tumors such as metastatic non-small cell lung cancer (NSCLC) and gastrointestinal cancers[2][3][5][8]. The approach aims to boost the efficacy of adoptive cell transfer immunotherapy by overcoming intrinsic resistance mechanisms within patient-derived T cells.
