Cisplatin + gefitinib is a combination therapy consisting of two small molecule drugs used primarily in oncology research and clinical trials. Cisplatin is a platinum-based chemotherapeutic agent that forms DNA crosslinks, leading to apoptosis in rapidly dividing cells. Gefitinib is an epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor that blocks EGFR signaling pathways involved in cell proliferation and survival. The combination has been studied for its potential to overcome resistance to cisplatin, particularly in non-small cell lung cancer (NSCLC) with wild-type or mutant EGFR, as well as other cancers such as urothelial carcinoma. Preclinical studies suggest the combination can enhance anti-tumor effects by promoting apoptosis and inhibiting proliferation more effectively than either drug alone[1][6]. However, some studies have reported antagonistic interactions when both drugs are administered simultaneously due to interference with drug uptake mechanisms[3]. The optimal sequencing and dosing of this combination remain under investigation.
