Overview

This combination therapy is a novel neoadjuvant regimen being evaluated for the treatment of locoregionally advanced head and neck squamous cell carcinoma (HNSCC). It consists of four distinct agents: **cisplatin**, a platinum-based DNA alkylating agent; **nab-paclitaxel**, an albumin-bound nanoparticle formulation of the microtubule inhibitor paclitaxel; **adebrelimab**, a monoclonal antibody targeting programmed death-ligand 1 (PD-L1); and **imaxitinib** (also known as SHR0302), a selective small-molecule inhibitor of Janus kinase 1 (JAK1). The regimen is designed to leverage the cytotoxic effects of chemotherapy to induce tumor cell death and antigen release, while adebrelimab restores T-cell mediated anti-tumor immunity. The addition of imaxitinib aims to modulate the tumor microenvironment and interferon-stimulated gene signatures to enhance the efficacy of the chemo-immunotherapy backbone. This specific combination is currently being investigated in a Phase 2 pilot trial led by the Second Affiliated Hospital, School of Medicine, Zhejiang University.
