This is a four-drug chemotherapy combination regimen consisting of cisplatin, 5-fluorouracil (also known as fluorouracil or 5-FU), vincristine, and doxorubicin. Each component has a distinct mechanism of action targeting rapidly dividing cancer cells: - **Cisplatin** is a platinum-based compound that forms DNA crosslinks and induces apoptosis by interfering with DNA replication and transcription. - **5-Fluorouracil** is an antimetabolite that inhibits thymidylate synthase, blocking DNA synthesis. - **Vincristine** is a vinca alkaloid that disrupts microtubule formation during mitosis. - **Doxorubicin** is an anthracycline antibiotic that intercalates into DNA and inhibits topoisomerase II. The combination has been studied primarily in pediatric oncology for the treatment of intermediate-risk unresectable hepatoblastoma (a type of liver cancer in children), where it demonstrated feasibility, tolerability, and efficacy. The addition of doxorubicin to the standard C5V regimen (cisplatin/fluorouracil/vincristine) was shown to intensify therapy for advanced disease[1][2][3]. This regimen may also be used or studied in other solid tumors.
