Overview

A combination chemotherapy regimen consisting of cisplatin, carboplatin, and paclitaxel, specifically utilized as adjuvant therapy in the treatment of high-risk endometrial cancer. This regimen, often referred to as the PORTEC-3 schedule, involves the administration of cisplatin concurrently with radiotherapy, followed by adjuvant cycles of carboplatin and paclitaxel. Cisplatin and carboplatin are platinum-based agents that induce DNA damage through the formation of intra-strand and inter-strand cross-links, inhibiting DNA replication and transcription. Paclitaxel is a taxane that binds to the beta-subunit of tubulin, promoting microtubule assembly and preventing disassembly, which leads to mitotic arrest and apoptosis. This combination is being evaluated within the RAINBO umbrella trial led by the Leiden University Medical Center to refine treatment based on molecular profiles.
