Overview

Cisplatin + dacarbazine is a combination chemotherapy regimen consisting of two small molecule cytotoxic agents—cisplatin (a platinum-based compound) and dacarbazine (an alkylating agent). This combination is primarily used in the treatment of metastatic melanoma. Cisplatin forms DNA crosslinks that inhibit DNA synthesis and function, leading to apoptosis in rapidly dividing cells. Dacarbazine acts as an alkylating agent that methylates guanine bases in DNA, resulting in inhibition of DNA replication and cell death. The regimen has demonstrated response rates between 10% and 55% in metastatic melanoma patients, with some studies reporting complete responses[1]. Both drugs are approved for use individually but are also commonly combined for synergistic antineoplastic effects.
