Overview

The combination of cisplatin, docetaxel, and mitomycin C was investigated as a treatment regimen for non-small cell lung cancer (NSCLC). This three-drug combination underwent a Phase I clinical trial to determine its maximum tolerated dose (MTD) and safety profile. Cisplatin is a platinum-based compound that works by crosslinking DNA, interfering with cell division and triggering apoptosis in cancer cells. Docetaxel is a taxane derivative that binds to microtubules, stabilizing them and preventing their disassembly, which disrupts cell division. Mitomycin C is an alkylating agent that crosslinks DNA and produces free radicals, causing DNA damage and inhibiting DNA synthesis. The study fixed docetaxel at 60 mg/m² and cisplatin at 80 mg/m², while escalating mitomycin C from 4 mg/m² to 6 mg/m². All drugs were administered on day 1 and repeated every 3-4 weeks. However, the addition of mitomycin C resulted in relatively high toxicities, with dose-limiting toxicities (DLTs) observed at the 6 mg/m² mitomycin C dose level. The MTD was determined to be 60 mg/m² for docetaxel, 80 mg/m² for cisplatin, and 6 mg/m² for mitomycin C. Due to these toxicities, further evaluation of this specific three-drug combination was deemed unwarranted.
