This is a combination regimen consisting of three agents: - **Cisplatin** (a platinum-based chemotherapeutic that forms DNA crosslinks, leading to apoptosis in rapidly dividing cells) - **Doxorubicin** (an anthracycline antibiotic that intercalates DNA and inhibits topoisomerase II, causing DNA damage and cell death) - **Interferon alpha-2b** (a recombinant cytokine with immunomodulatory, antiproliferative, and antiviral properties). The combination has been studied as a chemoimmunotherapy approach for several cancers. In malignant pleural mesothelioma, this regimen demonstrated an overall response rate of 29% in a phase II trial but was associated with significant toxicity, particularly myelosuppression and fatigue[1][8]. Preclinical studies suggest interferon alpha-2b may enhance the cytotoxicity of cisplatin. The combination has also been explored in other solid tumors such as hepatocellular carcinoma and biliary tract cancers[2][3][5].
