Cisplatin + etanidazole is an investigational combination of two small molecule drugs used primarily in oncology research. Cisplatin is a platinum-based alkylating agent that exerts its antineoplastic effects by forming DNA cross-links, thereby inhibiting DNA replication and transcription and inducing cell death. Etanidazole (SR-2508) is a third-generation 2-nitroimidazole compound developed as a hypoxic cell radiosensitizer; it enhances the cytotoxicity of radiation and certain chemotherapeutic agents by sensitizing hypoxic tumor cells to their effects. The combination has been studied in preclinical models and early-phase clinical trials for various cancers, including esophageal cancer, superficial tumors, and small-cell lung cancer. Studies suggest that adding etanidazole to cisplatin can increase tumor cell kill rates without proportionally increasing toxicity; however, renal function monitoring is important due to potential nephrotoxicity from both agents[1][6][9].
