Overview

This is a combination chemotherapy regimen consisting of three cytotoxic agents—cisplatin, fluorouracil (also known as 5-fluorouracil or 5-FU), and mitomycin C. Each drug has a distinct mechanism of action: - **Cisplatin** is a platinum-based compound that forms DNA crosslinks, leading to apoptosis in rapidly dividing cells. - **Fluorouracil** is an antimetabolite that inhibits thymidylate synthase, disrupting DNA synthesis and function. - **Mitomycin C** acts as an alkylating agent causing DNA crosslinking and strand breakage. This combination has been studied primarily for advanced or recurrent head and neck squamous cell carcinoma (HNSCC) and also in anal canal carcinoma. The regimen demonstrates high response rates but with notable hematologic toxicity. It may be used alone or alongside radiotherapy depending on the indication[1][2][4].
