A combination therapy consisting of cisplatin (a platinum-based chemotherapy), ganetespib (a heat shock protein 90 inhibitor), and pemetrexed (an antifolate antineoplastic agent). This triplet therapy works through multiple mechanisms: cisplatin causes DNA damage and prevents cancer cell replication, ganetespib blocks Hsp90-mediated signaling that cancer cells use to divide and grow while potentially enhancing T-cell-mediated antitumor immune responses, and pemetrexed stops cancer cells from making and repairing DNA by inhibiting folate-dependent enzymes. The combination has been studied primarily for malignant pleural mesothelioma in clinical trials such as MESO-02.
