Overview

CCRT + GP is a sequential treatment regimen used for the management of locoregionally advanced nasopharyngeal carcinoma (NPC), particularly in patients with N2-3 stage disease. The regimen consists of concurrent chemoradiotherapy (CCRT), which involves the administration of radiation therapy concurrently with cisplatin, followed by adjuvant chemotherapy with the gemcitabine and cisplatin (GP) doublet. Gemcitabine is a nucleoside analog antimetabolite that inhibits DNA synthesis by targeting ribonucleotide reductase and DNA polymerase, while cisplatin is a platinum-based cytotoxic agent that forms DNA adducts and crosslinks. This specific sequence was established as a potential standard of care following a Phase 3 clinical trial conducted by Sun Yat-sen University Cancer Center, which demonstrated improved locoregional control and survival outcomes compared to the traditional cisplatin and fluorouracil (PF) adjuvant approach.
