Overview

This is a combination regimen consisting of three agents: - **Cisplatin** (a platinum-based chemotherapy agent that forms DNA crosslinks and induces apoptosis in rapidly dividing cells) - **Gemcitabine** (a nucleoside analog that inhibits DNA synthesis by incorporation into DNA and inhibition of ribonucleotide reductase) - **Trastuzumab** (a humanized monoclonal antibody targeting the human epidermal growth factor receptor 2 [HER2], blocking HER2-mediated signaling and inducing antibody-dependent cellular cytotoxicity). The combination is used primarily for the treatment of HER2-positive cancers, notably in advanced biliary tract cancer (BTC) with HER2 overexpression. In recent phase II clinical trials, this regimen demonstrated improved progression-free survival compared to historical controls treated with chemotherapy alone. The combination has also been studied in other HER2-positive solid tumors such as non-small cell lung cancer and breast cancer. Trastuzumab specifically targets tumors overexpressing the HER2 protein, while cisplatin and gemcitabine provide broad cytotoxic effects[1][4][7].
