This is a combination regimen consisting of three drugs—cisplatin, imatinib mesylate, and pemetrexed—used in investigational cancer therapy. Cisplatin is a platinum-based chemotherapeutic agent that induces DNA crosslinking and apoptosis in rapidly dividing cells. Pemetrexed is an antifolate chemotherapy drug that inhibits several folate-dependent enzymes involved in nucleotide synthesis, thereby blocking DNA replication and cell division. Imatinib mesylate is a small molecule tyrosine kinase inhibitor targeting the platelet-derived growth factor receptor (PDGFR), BCR-ABL, and c-KIT kinases; it disrupts signaling pathways critical for tumor cell proliferation and survival. This combination has been studied primarily for unresectable malignant pleural mesothelioma (MPM) with the hypothesis that PDGFR pathway inhibition by imatinib may enhance the efficacy of standard cisplatin-pemetrexed chemotherapy[1][2][3][4].
