Overview

This combination chemotherapy regimen, consisting of cisplatin, paclitaxel, and gemcitabine, was developed and evaluated by the GERCOR (Multidisciplinary Oncology Cooperative Group) for the treatment of advanced urothelial carcinoma of the bladder. The regimen combines three agents with distinct mechanisms: gemcitabine, a nucleoside analog that inhibits DNA synthesis; cisplatin, a platinum-based agent that induces DNA cross-linking; and paclitaxel, a taxane that stabilizes microtubules to prevent mitosis. This triplet combination was specifically studied in a Phase II clinical trial (NCT00006118) to assess its efficacy in patients with progressive unresectable regional or metastatic bladder cancer, aiming to improve response rates through synergistic cytotoxic effects.
