Overview

Cisplatin is a platinum-based chemotherapy drug classified as an alkylating agent. It works by binding to and cross-linking DNA, forming DNA adducts that disrupt DNA replication and transcription, ultimately leading to cell cycle arrest and apoptosis in rapidly dividing cells such as cancer cells[1][4][5][6]. Cisplatin is used alone or in combination with other therapies—including radiotherapy—to treat various solid tumors, including cancers of the bladder, ovary, testicle, head and neck, lung, cervix, esophagus, brain (including neuroblastoma), mesothelioma, sarcomas, lymphomas, and germ cell tumors[2][3][4][5]. When combined with radiotherapy (cisplatin + radiotherapy), it acts as a radiosensitizer—enhancing the effect of radiation on tumor cells. The combination is commonly used for locally advanced cancers such as head and neck squamous cell carcinoma or cervical cancer.
