Overview

This is a combination chemotherapy regimen consisting of three cytotoxic agents—cisplatin, vinorelbine, and gemcitabine. It is primarily used in the treatment of advanced non-small cell lung cancer (NSCLC), particularly as first-line therapy for patients with stage IIIB or IV disease. The combination leverages the distinct mechanisms of action of each agent to maximize antitumor efficacy while maintaining a manageable toxicity profile. Clinical studies have demonstrated significant response rates and survival benefits in NSCLC patients treated with this regimen[1][3][8]. Cisplatin acts as a DNA crosslinker causing apoptosis; vinorelbine inhibits microtubule assembly leading to mitotic arrest; and gemcitabine is a nucleoside analog that disrupts DNA synthesis during the S-phase[6]. This triplet regimen has been evaluated in multiple phase II trials for its efficacy and safety.
