Cisplatin-albumin complex is a **formulation of cisplatin in which the drug is covalently or non-covalently bound to human serum albumin (HSA)**. This complex serves as a nanocarrier for cisplatin with potential advantages in targeted drug delivery and reduced nephrotoxicity. The therapeutic efficacy is believed to be enhanced in tumors expressing the albumin-binding protein SPARC, enabling both passive (by EPR effect) and active targeting to tumor tissue. Mechanistically, the complex’s anticancer activity derives from the **release of active cisplatin**, which then cross-links DNA in tumor cells, causing cell cycle arrest and apoptosis. Preclinical and clinical studies indicate the complex can achieve comparable tumor growth suppression as free cisplatin, with improved safety—particularly a marked reduction in nephrotoxicity and lower unbound plasma platinum. Cisplatin-albumin complex was investigated in phase I clinical trials for squamous cell carcinoma of the head and neck and in preclinical glioblastoma models, showing the possibility of expanded use for SPARC-expressing tumors[2][3][4].
