Cisplatin liposomal is a synthetic formulation of cisplatin encapsulated within a lipid bilayer to form nanoparticles. This encapsulation enhances tumor bioavailability and reduces toxicity compared to conventional cisplatin. The most studied formulation is Lipoplatin, which has shown similar or superior efficacy with reduced nephrotoxicity in clinical trials. The mechanism involves enhanced delivery of cisplatin into tumor cells via fusion or endocytosis of the lipid nanoparticle, leading to DNA crosslinking and apoptosis. It is primarily developed for solid tumors like non-small cell lung cancer (NSCLC) and pancreatic cancer, allowing higher platinum concentrations in tumors while sparing normal tissues.
