A citrullinated enolase 1 (ENO1) vaccine is a peptide-based cancer vaccine designed to elicit an immune response against citrullinated forms of the enzyme enolase 1, which becomes immunogenic due to abnormal protein modification (citrullination) observed in cancer cells. The vaccine’s mechanism relies on activating both CD4+ and CD8+ T cells against tumor cells presenting citrullinated ENO1 epitopes via MHC class II. This triggers pro-inflammatory cytokine release and direct cytotoxicity of tumor cells, leading to tumor growth delay and improved survival in preclinical models. In clinical and preclinical trials, such vaccines have shown immune activation, delayed tumor progression, and enhanced efficacy when combined with other immunotherapies (such as checkpoint inhibitors). The vaccine is under development for multiple solid tumors including triple-negative breast cancer, head and neck cancer, ovarian cancer, and renal cancer[1][2][3][4][5].
