Overview

This is a novel cell therapy approach utilizing Chimeric Antigen Receptor (CAR) engineered regulatory T cells (Tregs) designed to target citrullinated vimentin (CV). Citrullinated vimentin is a post-translationally modified protein found in the extracellular matrix of inflamed tissues, particularly in autoimmune conditions like rheumatoid arthritis (RA). The CAR-Tregs are engineered to specifically recognize and bind to CV, leading to their activation and expansion at the site of inflammation. Once activated, these CAR-Tregs are intended to suppress local immune responses, re-establish immune tolerance, and reduce inflammation, offering a targeted therapeutic strategy for autoimmune diseases. Initial development involved researchers from institutions like UCSF and the Institute for Research in Biomedicine, with companies like Sonoma Biotherapeutics advancing similar concepts into clinical development for rheumatoid arthritis.
