**CiVi 008** is a third-generation, long-acting oral antisense oligonucleotide targeting PCSK9, developed by CiVi Biopharma for hypercholesterolemia and atherosclerotic cardiovascular disease (ASCVD) prevention. It inhibits PCSK9 synthesis at the mRNA level using locked nucleic acid (LNA) technology with GalNAc conjugation for liver-specific delivery, aiming for >60% stable LDL-C reduction comparable to monoclonal antibodies but with daily oral dosing and lower cost of goods sold. Positioned as the only oral PCSK9-targeting ASO in development, it leverages a minor pathway for LDL degradation distinct from approved anti-PCSK9 mAbs.[1][4][5][11]
