Drug intelligence / Profile preview
This is an investigational **combination therapy** evaluated in phase II clinical trials for metastatic castration-resistant prostate cancer (mCRPC) in patients previously treated with docetaxel. The regimen includes: - **Cixutumumab**: A fully human IgG1 monoclonal antibody targeting the insulin-like growth factor 1 receptor (IGF-1R), leading to receptor internalization, degradation, and inhibition of IGF-1R-mediated signaling. This may also induce antibody-dependent cellular cytotoxicity. Developed initially by ImClone Systems (later acquired by Eli Lilly), cixutumumab modulates growth and survival pathways crucial to tumor cell proliferation[1][4][5][8]. - **Mitoxantrone**: A synthetic anthracenedione antineoplastic agent that intercalates into DNA, inhibiting topoisomerase II and leading to DNA strand breaks and decreased DNA/RNA synthesis. It is approved in combination with corticosteroids for hormone-refractory prostate cancer pain and has broad antineoplastic activities[1][7]. - **Prednisone**: A synthetic glucocorticoid used for its anti-inflammatory and immunosuppressive effects; in oncology, it is commonly used as supportive therapy in combination with cytotoxic agents[1]. This combination has shown feasibility and moderate toxicity in phase II trials for mCRPC, with cixutumumab's addition hypothesized to enhance the antitumor activity of mitoxantrone and prednisone. The most common adverse event was fatigue. The regimen is not currently approved or marketed for this indication[1][4][6].
