CJC-1131 is a **long-acting, albumin-conjugated glucagon-like peptide 1 (GLP-1) analogue** resistant to dipeptidyl peptidase-IV (DPP-IV) degradation, designed to prolong the half-life and therapeutic activity of GLP-1. CJC-1131 couples covalently to endogenous human serum albumin via a reactive linker at the C-terminus, which dramatically extends its plasma half-life (to 8.9–14.7 days in humans) compared to native GLP-1. CJC-1131 selectively activates the glucagon-like peptide 1 receptor (GLP-1R), leading to enhanced insulin secretion, improved glucose tolerance, and potential islet neogenesis in preclinical models. It was developed as a potential therapy for type 2 diabetes mellitus, showing reductions in HbA1c and fasting glucose without severe hypoglycemia. CJC-1131 underwent Phase 2 clinical trials but is **no longer in active development**. It was originally developed by Conjuchem.
