Overview

Cjoc42 is a small molecule inhibitor of PSMD10 (also known as gankyrin), an oncogenic protein that serves as a non-ATPase subunit of the 26S proteasome. PSMD10 is frequently overexpressed in multiple myeloma (MM) and other malignancies, where it acts as a selective suppressor of MHC class II (MHC-II) expression, contributing to an immunosuppressive tumor microenvironment and impaired CD4+ T-cell activation. By inhibiting PSMD10, Cjoc42 restores the expression of MHC-II components, such as HLA-DR and CD74, on the surface of malignant plasma cells. This restoration of antigen presentation pathways is designed to enhance anti-tumor immunity and potentially overcome resistance to standard therapies like proteasome inhibitors and immunomodulatory drugs (IMiDs).
