CJRB-101 is a live, novel therapeutic bacterial strain developed for its immunostimulating and antineoplastic (anti-cancer) activities. Upon administration, CJRB-101 activates M1 macrophages and induces the repolarization of tumor-promoting M2 macrophages into anti-tumor M1 macrophages, thereby enhancing immune-mediated tumor suppression. This mechanism increases cytotoxic T lymphocyte (CD8+ T cell) activity within the tumor microenvironment. The drug’s anti-cancer effects are mediated through Toll-like receptor 4 (TLR4)-dependent stimulation of key immune cell populations, including macrophages and CD8+ T cells. Preclinical studies have demonstrated efficacy in humanized mouse models of non-small cell lung cancer and other solid tumors. Clinical development is ongoing for advanced cancers such as non-small-cell lung cancer (NSCLC), head and neck squamous cell carcinoma (HNSCC), melanoma, and small cell lung cancer—primarily in combination with pembrolizumab to address resistance to immune checkpoint inhibitors.
