CK-0045

CK-0045 is a long-acting, lipidated analogue of interleukin-22 (IL-22), an atypical, non-immunomodulatory cytokine that selectively targets epithelial cells. It acts as an IL-22 agonist and is designed to enhance the pharmacologic properties of endogenous IL-22 for therapeutic use. CK-0045 is being developed primarily for the treatment of obesity and type 2 diabetes mellitus, with additional potential in conditions characterized by epithelial injury such as inflammatory bowel disease. The drug leverages lipidation technology to extend its circulation time in the body, enabling once-weekly subcutaneous administration. Clinical studies have shown exposure-dependent reductions in body weight and improvements in metabolic parameters such as insulin resistance and LDL cholesterol, with a favorable safety profile observed so far. CK-0045 was originally in-licensed from Novo Nordisk and is currently under development by Cytoki Pharma[1][3][4][6][7].
