CK-303 is a fully human monoclonal antibody targeting carbonic anhydrase IX (CAIX), a cell-surface enzyme that is highly expressed in over 85% of renal cell carcinoma (RCC) cases but has very limited expression in normal tissues. Developed by Checkpoint Therapeutics through a license from the Dana-Farber Cancer Institute, CK-303 is designed to bind specifically to CAIX-expressing tumor cells and induce cell death via antibody-dependent cell-mediated cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). The drug is currently in preclinical development for the treatment of RCC and other CAIX-positive solid tumors, with potential for use in combination with other immunotherapies such as anti-PD-L1 or anti-GITR antibodies.
