CK1α activators are a class of small molecule therapeutics being developed by StemSynergy Therapeutics to treat colorectal and other Wnt-driven cancers. These compounds function by activating Casein Kinase 1 alpha (CK1α), a key negative regulator of the Wnt/β-catenin signaling pathway. By enhancing CK1α activity, these activators promote the phosphorylation and subsequent degradation of β-catenin, thereby suppressing the aberrant Wnt signaling that drives tumor growth and survival in specific oncogenic contexts. The program is currently in the IND-enabling phase of development.
