Overview

CK2 shRNA is a research-grade short hairpin RNA (shRNA) construct used to genetically silence the expression of Casein Kinase 2 (CK2), a ubiquitous serine/threonine protein kinase that is frequently overexpressed in various cancers and inflammatory conditions. By targeting the mRNA of the CK2 catalytic subunits (specifically CK2α or CK2α', encoded by the CSNK2A1 and CSNK2A2 genes), the shRNA utilizes the RNA interference (RNAi) pathway to knock down protein expression. In preclinical oncology research, CK2 shRNA is employed to investigate the kinase's role in cell cycle progression, apoptosis evasion, and the regulation of oncogenic transcription factors like Ikaros. It is primarily used as a genetic validation tool to confirm the therapeutic potential of CK2 inhibition, often serving as a comparator for pharmacological inhibitors such as silmitasertib (CX-4945).
