Overview

CK37 is a small molecule inhibitor of choline kinase-alpha (ChoKα) being developed by Advanced Cancer Therapeutics. It functions by competitively inhibiting the enzyme responsible for the phosphorylation of choline to phosphocholine, a critical step in the Kennedy pathway for phosphatidylcholine synthesis. This disruption of phospholipid metabolism leads to decreased levels of phosphocholine and attenuates downstream survival signaling pathways, including MAPK and PI3K/AKT. Preclinical evidence suggests that CK37 selectively targets neoplastic cells and demonstrates anti-tumor activity in various models, including lung, breast, ovarian, brain, and prostate cancers.
