The CKAP2 modulator is a first-in-class, oral small molecule being developed by Soley Therapeutics for the treatment of various cancers. It targets Cytoskeleton-Associated Protein 2 (CKAP2), an intrinsically disordered protein and potent microtubule growth factor that is often overexpressed in malignant tissues and associated with poor prognosis. By modulating CKAP2, the drug disrupts the microtubule network and inhibits the FAK-ERK2 signaling pathway, leading to G2/M cell cycle arrest, reduced cell migration, and decreased angiogenesis. Preclinical studies have demonstrated significant anti-tumor activity in models of non-small cell lung cancer and colon cancer, as well as high sensitivity in large B cell lymphoma and acute myeloid leukemia cell lines.
