CKD-215 is an orally bioavailable small molecule inhibitor of poly(ADP-ribose) polymerase (PARP), developed by Chong Kun Dang Pharmaceutical. It is being developed as a generic or bioequivalent version of olaparib (Lynparza). The drug targets PARP1, PARP2, and PARP3, enzymes essential for repairing single-strand DNA breaks via the base excision repair pathway. In cancers with BRCA1 or BRCA2 mutations—which are already deficient in homologous recombination repair—PARP inhibition prevents the repair of DNA damage, leading to the accumulation of double-strand breaks that the cell cannot repair. This results in genomic instability and selective cell death, a mechanism known as synthetic lethality. CKD-215 is currently in Phase 1 clinical development for the treatment of advanced BRCA-mutated ovarian, fallopian tube, and primary peritoneal cancers.
