CKD-504 is a novel, selective small molecule inhibitor of histone deacetylase 6 (HDAC6), developed by Chong Kun Dang Pharmaceutical. It is being investigated primarily for the treatment of neurodegenerative diseases such as Huntington's disease and has also shown potential in preclinical models for Charcot-Marie-Tooth disease type 1A (CMT1A). The drug works by inhibiting HDAC6 activity, leading to increased acetylation of α-tubulin and stabilization of microtubules. This results in improved axonal transport, reduced accumulation of mutant huntingtin protein (mHTT), decreased inflammation and tau hyperphosphorylation in Huntington’s disease models. In CMT1A models, CKD-504 reduces PMP22 protein levels and restores myelination. The compound is currently in Phase 1 clinical trials for Huntington's disease[1][2][4][5][6][7].
