Overview

CKD-519 is a potent, small-molecule cholesteryl ester transfer protein (CETP) inhibitor developed by Chong Kun Dang Pharmaceutical for the treatment of dyslipidemia. It works by inhibiting the CETP-mediated transfer of cholesteryl esters from high-density lipoprotein (HDL) to low-density lipoprotein (LDL) and very-low-density lipoprotein (VLDL), which results in a significant increase in HDL cholesterol (HDL-C) levels. Classified as a BCS Class II drug, CKD-519 is characterized by low aqueous solubility and high permeability, with high plasma protein binding. Clinical development has progressed through Phase II trials, where the drug has been evaluated both as a monotherapy and in combination with HMG-CoA reductase inhibitors (statins) such as atorvastatin and rosuvastatin. Studies have demonstrated that CKD-519 can achieve substantial inhibition of CETP activity and is suitable for once-daily oral administration.
