Overview

CL 316243 is a potent and highly selective β3-adrenoceptor (beta-3 adrenergic receptor) agonist. It was developed as a synthetic small molecule for research purposes and has demonstrated significant anti-obesity and anti-diabetic effects in preclinical models. The drug increases brown adipose tissue thermogenesis and metabolic rate by stimulating lipolysis in adipocytes. This leads to increased energy expenditure, reduced blood glucose levels (via enhanced insulin secretion), improved insulin sensitivity, and decreased adiposity. Its selectivity for the β3-adrenoceptor is over 10,000-fold greater than for β1 or β2 receptors[1][3][5]. While it has shown promise in animal studies for obesity and type 2 diabetes mellitus (non-insulin-dependent diabetes), as well as potential use in urge urinary incontinence due to its effects on smooth muscle relaxation[5], it remains an experimental compound not approved for human therapeutic use.
