Overview

CL04183 is a synthetic peptide-based therapeutic developed by Cleara Biotech. It is designed to selectively eliminate "scarred" senescent cells, a subtype of cellular senescence associated with late-stage cancer and certain chronic diseases. The drug acts as an inhibitor of the interaction between FOXO4 (forkhead box O4) and p53 (tumor protein p53), thereby stimulating apoptosis in targeted cells. This mechanism aims to counteract metastatic burden and infiltration in cancers such as metastatic colorectal cancer (mCRC) and triple-negative breast cancer (TNBC). Preclinical studies have shown that CL04183 has favorable pharmacokinetics, high plasma stability, resistance to liver enzyme degradation, and efficacy at doses well below its maximum tolerated dose[1][2][4][5].
