Clade C gp140 HIV bivalent vaccine (JNJ-65184340) is an investigational recombinant bivalent protein subunit vaccine developed by Janssen Vaccines & Prevention (a subsidiary of Johnson & Johnson) for the prevention of HIV-1 infection. The vaccine consists of two active components: a recombinant HIV-1 envelope glycoprotein gp140 of Clade C and a recombinant Mosaic gp140 engineered to represent multiple HIV-1 variants, both produced in the PER.C6 cell line and adjuvanted with aluminum phosphate. It was evaluated in combination with the viral vector vaccine Ad26.Mos4.HIV as a heterologous prime-boost regimen in the Phase 3 Mosaico efficacy trial (NCT03964415). In January 2023, the trial was discontinued after an independent data safety monitoring board determined that the regimen lacked efficacy in preventing HIV-1 infection compared to placebo, though no safety concerns were identified.
