claudin 18.2 targeted proapoptotic peptide is an experimental peptide-drug conjugate (PDC) designed for the targeted treatment and early diagnosis of pancreatic adenocarcinoma. The agent consists of a specific peptide ligand that binds to Claudin 18.2 (CLDN18.2), a tight junction protein that is highly expressed and accessible on the surface of pancreatic and gastric cancer cells due to the loss of cell polarity. This targeting peptide, identified through phage display screening, is conjugated to a proapoptotic peptide payload. Upon binding to the tumor cells, the conjugate induces apoptosis, leading to tumor cell death. Preclinical data from KPC transgenic mouse models of pancreatic cancer have shown that the peptide conjugate significantly accumulates in tumor tissues and improves overall survival, highlighting its potential as a precision medicine candidate for aggressive and metastatic pancreatic neoplasms.
