Claudin-4 (CLDN4) small interfering RNA (siRNA) is an RNA interference-based therapeutic approach designed to silence the expression of the CLDN4 gene. CLDN4 is a tight junction protein frequently overexpressed in various cancers, including bladder urothelial carcinoma (BUC), where it correlates with cancer progression, stemness, and drug resistance. By targeting CLDN4 mRNA for degradation, the siRNA reduces the levels of both tight-junction-integrated and unintegrated CLDN4 protein. Research indicates that reducing CLDN4 levels can inhibit cell proliferation, decrease metastatic potential, and sensitize cancer cells to apoptosis, potentially through the modulation of FAK phosphorylation and integrin β1 signaling. While primarily used as a research tool to investigate the oncogenic mechanisms of CLDN4, it represents a potential therapeutic strategy for cancers overexpressing this protein.
