Claudin-6 ADC refers to a class of antibody-drug conjugates designed to target Claudin-6 (CLDN6), a tight junction protein that is highly expressed in various solid tumors—including ovarian, testicular, endometrial, and gastric cancers—but largely absent in healthy adult tissues. A specific novel candidate described by ADC Therapeutics and Myricx Bio utilizes a humanized IgG1 antibody directed against CLDN6, site-specifically conjugated to a camptothecin payload via a cleavable valine-alanine linker. This ADC features a drug-to-antibody ratio (DAR) of 6 and incorporates Fc mutations to abrogate FcγR-mediated effector functions, thereby reducing potential off-target toxicity. Preclinical studies presented at AACR 2024 demonstrated potent cytotoxicity, specific anti-tumor activity, and bystander killing in CLDN6-expressing cell lines and patient-derived xenograft (PDX) models, with favorable tolerability observed in non-human primate studies.
