Claudin18.2-targeted CAR-T cells are an autologous cell therapy consisting of T cells genetically engineered to express a chimeric antigen receptor (CAR) that specifically recognizes Claudin 18.2 (CLDN18.2). CLDN18.2 is a tight junction protein whose expression is normally confined to the gastric mucosa but is frequently upregulated in various gastrointestinal malignancies, including gastric, gastroesophageal junction, and pancreatic adenocarcinomas. This restricted expression pattern makes it an ideal target for precision immunotherapy. Once infused, these CAR-T cells bind to CLDN18.2 on the surface of tumor cells, triggering T-cell activation and the release of cytotoxic granules to induce tumor cell death. In clinical trials conducted by Hainan Cancer Hospital, these cells are being evaluated in combination with Claudin18.2-targeted activated dendritic cells to treat advanced pancreatic cancer, aiming to enhance T-cell infiltration and persistence within the tumor microenvironment.
