Overview

Claudin18.2-targeting CAR-T cells are a class of autologous or allogeneic chimeric antigen receptor (CAR) T-cell therapies engineered to target Claudin 18.2 (CLDN18.2), a tight junction protein isoform frequently overexpressed in gastrointestinal cancers such as gastric, gastroesophageal junction, and pancreatic adenocarcinomas. These therapies involve the genetic modification of T cells to express a CAR that recognizes CLDN18.2, leading to targeted tumor cell lysis. Several candidates are in clinical development, most notably satricabtagene autoleucel (CT041), which has demonstrated significant efficacy in refractory gastrointestinal cancers. Other variations include armored CAR-Ts like AZD6422, which incorporate modifications such as TGF-β receptor modifications to overcome the immunosuppressive tumor microenvironment.
